LIPID A and BACTERIAL LIPOPOLYSACCHARIDES

چکیده
1. Structure and Occurrence The envelope of Gram-negative bacteria, including those of human pathogens such as Escherichia coli and Salmonella enterica, is composed of two distinct lipid membranes: an inner membrane, consisting of glycerophospholipids, and a highly distinctive outer membrane. The latter is an asymmetric bilayer, the outer leaflet of which in most species consists predominantly of lipopolysaccharides of which Lipid A is a key component. They are of great scientific interest in that they are toxins and stimulate strongly the innate immune system in eukaryotic host species. Proteins take up much of the remaining surface of the outer leaflet of the outer membrane, while the inner leaflet is composed of conventional glycerophospholipids, mainly phosphatidylethanolamine, phosphatidylglycerol and cardiolipin.

Macrophage stimulation by bacterial lipopolysaccharides. II. Evidence for differentiation signals delivered by lipid A and by a protein rich fraction of lipopolysaccharides
Stimulation of macrophages to lyse tumor cells is a property common to lipopolysaccharide (LPS) extracted from a variety of smooth and rough bacterial strains by several different preparative procedures. The relationship between macrophage stimulation and the structural characteristics of LPS is defined. In protein-free LPS, lipid A bears the stimulatory signal which results in the differentiat...
متن کاملLipid A as the biologically active moiety in bacterial endotoxin (LPS)-initiated generation of procoagulant activity by peripheral blood leukocytes.
Preparations of rabbit or human leukocytes, when incubated with bacterial endotoxins (lipopolysaccharides, LPS) are stimulated to generate a procoagulant-tissue factor activity (TFa). As LPS has been shown to consist of specific repeating oligosaccharide side chains (O-antigen) linked to a central polysaccharide core region that is, in turn, linked to the lipid region of the molecule (lipid A),...
متن کاملPyrogenicity and immunogenicity of lipid A complexed with bovine serum albumin or human serum albumin.
The lipid A component of bacterial lipopolysaccharides (endotoxins), when complexed to bovine serum albumin (BSA) or human serum albumin (HSA), was shown to be a potent pyrogen. Furthermore, rabbits could be protected against endotoxin fever by immunization with both lipid A.BSA and lipid A.HSA complexes. The results presented in this paper show that lipid A is responsible for the pyrogenic act...
متن کاملA rapid, small-scale procedure for the structural characterization of lipid A applied to Citrobacter and Bordetella strains: discovery of a new structural element.
Endotoxins [lipopolysaccharides (LPSs)] are part of the outer cell membrane of Gram-negative bacteria. Their biological activities are associated mainly with the lipid component (lipid A) and even more specifically with discrete aspects of their fine structure. The need for a rapid and small-scale analysis of lipid A motivated us to develop a procedure that combines direct isolation of lipids A...
